Abstract
Over the past two decades, researchers have developed various culture strategies to enhance the lutein content of algae biomass. However, the lutein content does not yet meet requirements for commercialization. An important research topic is determining the relationships between lutein and other key metabolic compounds under different culture conditions. Such findings will provide insights for developing new strategies to enhance lutein content. According to the functionality of lutein in higher plants and the fact that lutein shares same precursors with chlorophyll in the metabolic pathway, lutein is expected to show a close correlation with chlorophyll. In addition, nitrogen concentration and different light intensities may also affect this correlation. However, the effect of these factors on the correlation of lutein and chlorophyll content in Chlorella Sorokiniana has not yet been identified. In this study, several experiments were designed and conducted to investigate these effects. The results showed a strong correlation between lutein and chlorophyll biosynthesis under high [N] conditions. Under low [N] conditions, both lutein and chlorophyll contents were significantly lower than high [N] condition. In addition, the correlation between lutein and chlorophyll content significantly altered under low [N] conditions. Furthermore, compared with heterotrophic cultivation, mixotrophic cultivation under gentle light irradiance increased both lutein and chlorophyll contents. However, it had no significant effect on the lutein/chlorophyll ratio in high [N] condition. Moreover, harsh irradiance was found to increase the contents of the two pigments and to change lutein/chlorophyll ratio in low [N] conditions. However it had no significant effects in high [N] conditions.
