Effects of a Semen Extender Treated with Coenzyme Q10-Ubiquinol on the Quality of Thawed Ram Semen

The study aimed to explore whether the addition of 150μmol/L Coenzyme Q10 - ubiquinol(treatment) to a glycerol extender improved the quality of ram semen cryopreserved when compared to the results for the same extender without the addition of ubiquinol (control). Semen from 6 mature Suffolk rams was collected by electroejaculation on 6 occasions during the breeding season and the semen from each collection was pooled. Aliquots of the pooled semen were processed and frozen then thawed following the addition of the control or ubiquinol-treated extender. Computer assisted sperm analysis (CASA) evaluated the sperm motility and kinetic parameters. Plasma membrane integrity was determined a measure of viability. Measurements were made 10, 20, 60 and 120 min after thawing. Progressive motility was higher in ubiquinol- treated semen samples (P < 0.0001). Curvilinear velocity (P =0.002) straight line velocity (P =0.003) and time average velocity (P =0.001) were higher in the samples cryopreserved with the ubiquinol-treated extender. Spermatozoa tended to be more viable in samples in which the extender was treated with ubiquinol (P = 0.07). There were time-related decreases in the values of all variables regardless of whether they had the control or treated extender added. Ubiquinol improved the quality of cryopreserved ram semen by improving progressive motility and sperm kinetics and tended to also increase viability.
